Calculate the Dilution Required To Prepare a Working Solution of Your Antibody

The dilution calculator is a useful tool which allows you to calculate how to dilute a stock solution of antibody to a known concentration. Enter C1, C2 and V2 to calculate V1.

The Dilution Calculator Equation

The dilution calculator is based on the following equation:

Concentration(start) x Volume(start) = Concentration(final) x Volume(final)

This is the C1V1 = C2V2 equation.

An Example of a Dilution Calculation Using the Dilution Calculator

You received an antibody at a stock concentration of 10 mg/ml. You have five mice to inject with 200 µg per mouse. You want to inject each mouse with a 200 µL injection volume. This means you will need 1,000 µg in 1,000 µL (1 milliliter of a 1 mg/ml solution).

Using the equation C1V1 = C2V2, where C1= 10 mg/ml, C2= 1 mg/ml, V2= 1 ml and V1 is the unknown:

  1. Enter 10 in the Concentration (start) box and select the correct unit (mg/ml).
  2. Enter 1 in the Concentration (final) box and select the correct unit (mg/ml).
  3. Enter 1,000 in the Volume (final) box and select the correct unit (microliter).
  4. Press calculate.
  5. The answer of 100 microliter (0.1 ml) appears in the Volume (start) box.

How do I dilute my antibody to a working concentration?

We recommend diluting our antibodies in the InVivoPure dilution buffer matching the pH that the antibody is supplied in. The recommended InVivoPure dilution buffer for each antibody is listed on each product page. In the absence of the recommended InVivoPure dilution buffer, sterile 1x PBS matching the exact pH of the antibody formulation can be used. It is important to match the pH of the dilution buffer to the antibody solution to avoid aggregation.

When diluting we recommend using cold buffer and maintaining sterility by working in a biological safety cabinet and using sterile pipet tips, tubes, syringes, and buffers.

FAQs

How do you calculate how much to dilute? ›

To make a fixed amount of a dilute solution from a stock solution, you can use the formula: C1V1 = C2V2 where: V1 = Volume of stock solution needed to make the new solution. C1 = Concentration of stock solution. V2 = Final volume of new solution.

How do you calculate 1 in 10 dilution? ›

Commonly used dilutions are 1:10 or 1:2. Note that this is expressed as the ratio of the previous solution to the final volume of the dilution. For example, to make a 1:10 dilution, you add 1ml of your solution to 9ml of diluent for a final volume of 10ml.

What is a 1 to 20 dilution? ›

A 1:20 dilution implies that you take 1 part of stock solution and add 19 parts of water to get a total volume of diluted solution equal to 20 times that of the stock solution.

How much is 5 to 1 dilution? ›

Answer: 1:5 dilution = 1/5 dilution = 1 part sample and 4 parts diluent in a total of 5 parts. If you need 10 ml, final volume, then you need 1/5 of 10 ml = 2 ml sample. To bring this 2 ml sample up to a total volume of 10 ml, you must add 10 ml - 2 ml = 8 ml diluent.

How to do dilution formula? ›

You normally use the formula C1V1 = C2V2 to calculate dilutions: C1 is the concentrated starting stock. V1 is the volume of starting stock required. C2 is the desired stock concentration.

How to calculate dilution ratio? ›

The procedure is straightforward: add the two numbers together and divide the total volume by that sum. For example, if your dilution ratio is 10 to 1 and you want to fill a 12-ounce bottle, you would add 10 and 1 to get 11. Then, divide 12 by 11 to calculate the number of ounces required for the dilution.

How to do simple dilution? ›

Simple Dilution (Dilution Factor Method)..

For example, a 1:5 dilution (verbalize as "1 to 5" dilution) entails combining 1 unit volume of diluent (the material to be diluted) + 4 unit volumes of the solvent medium (hence, 1 + 4 = 5 = dilution factor).

How do you mix a 1 to 10 ratio? ›

A ratio of 1:10 means add 1 part of product to 10 parts of water. A ratio of 1:25 means add 1 part of product to 25 parts of water. 10 capfuls of water. You can use a measuring jug and convert the ratio to units of liquid measure.

How to do a 1 to 25 dilution? ›

Answer and Explanation:

We wish to dilute the 1M solution 25 times. We wish for this new solution to have a volume of 50ml. Hence, 1 out of 25 'units' of new solution (where each unit is equal to 50ml / 25 = 2ml) is made from the original 1M solution. The remaining 24 units are simply water.

How does a 1 10 dilution work? ›

To complete a tenfold dilution, the ratio must be 1:10. The 1 represents the amount of sample added. The 10 represents the total size of the final sample. For example, a sample size of 1 ml is added to 9 ml of diluent to equal a total of 10 ml.

How do I calculate dilution factor? ›

The formula for dilution factor (or DF for short) is as follows: DF = (final volume of cells + stain)/(initial volume of cells). For example, If you mix your sample 1:1 with AO/PI, you'll need to add 20 uL AO/PI to 20 uL cells, for a total of 40 uL.

What does 1 part to 20 parts water mean? ›

Example: The suggested dilution ratio 20:1 means 1 part of chemical product for every 20 parts of water. The same dilution ratio could be written as 1:20 meaning 20 parts of water to 1 part of chemical product.

How to do a 10 to 1 dilution? ›

For example, a 10:1 ratio means you mix 10 parts water to 1 part chemical.
