What’s The Right Number: Passaging Primary Cells | Kosheeka (1)

Primary cell cultures represent the physiological state of the tissue of origin. However, they have a finite lifespan undergo senescence after a certain number of passages or subcultures. While lower passages of the cells usually contain a mixed cell population, high passages can lead to loss of the differentiated properties of the cell type. Continuous culturing of primary cells can lead to accumulation of genetic and phenotypic changes in cells thus losing their original properties. There are multiple factors that can drive these changes including selective pressure of the culture media and growth conditions. Moreover, multiple passages can increase susceptibility of the primary culture to microbial contamination. Generally for primary cultures, passaging primary cells in between 2 to 5 are suitable for maintaining the genetic and phenotypic properties.

How To Avoid Increased Passaging Primary Cells Numbers

To access your primary culture at low passages, it is recommended to freeze down multiple vials of your stock cultures. This ensures that your experiments can be repeated with reproducibility at similar passages. Kosheeka offers high quality and pure population of primary cells at very low passages for your effective research. Our cells have undergone strict quality control and microbiological testing to ensure high reliability. In general, it is a healthy practice to passage a minimum number of times as per your experimental requirements and freeze down your working culture vials for future requirements. Seeding density is also important to prevent slow or overgrowth of cells.

A very low density can often be an additional stress to the primary cells while a high density will make passaging very frequent. Finally, one must record the passage number correctly and label the flasks after every passage to avoid confusion. There is no way one can determine the passage number of cells experimentally. However, for freezing the cells, the passage number must not be increased.

How do you know if a passage is no longer needed?

Primary cells from different patients respond differently to cell culture. Therefore, it is essential to monitor the morphology routinely and observe for any abnormalities or changes under an inverted microscope. It also recommended to characterize the cells at various passages and determine their growth curves to establish population doubling time.

Is passage number the definite guide to the age of cells?

Passage number does not tell us the actual number of population doublings. Depending on the seeding density of the cells, the number of cell divisions will vary. If the split ratio is 1:2, cells will undergo fewer doublings as compared to flasks with a split ratio of 1:5. So one must not rely solely on the passage number to assess other properties of cells. Passage number must only be used for reference purpose only.

In biology, a subculture is either a new cell culture or a microbiological culture made by transferring some or all cells from a previous culture to fresh growth medium. This action is called subculturing or passaging the cells.

What is the passage number for primary cells? ›

It is advisable to use primary cells below passage 10 for invitro studies. If you wish to use established cell line for invitro studies, try to use cells below passage 20-25.

How many cells should I passage? ›

Although they can be cultured for very long periods of time, most researchers and laboratories have guidelines as to how many passages they would use cells before they thaw a new vial (most guidelines suggest that scientists will use cells unto 50-75 passages before they will retire those cells and thaw a new vial).

How many cell passages is too much? ›

Practically speaking, cell passages should be limited to prevent population and genetic drift, and ideally, experiments performed with similar passage numbers. There is no defined maximum number of passages you should perform for a cell line, but it is highly recommended to keep passage numbers low.

What is the limit of cell passage numbers? ›

For the majority of cell cultures in the ECACC catalogues the limit of the number of passages they can undergo has not been determined. Finite cell lines derived from primary cells will senesce after a number of PDs characteristic of the cell type (e.g. for normal human fibroblasts this is around 60 PDs).

How many passages are A549 cells good for? ›

To ensure maximum efficiency, do not passage A549-ASC cells more than 20 times. Quality Control • The induction of ASC::GFP expression has been verified by Western blot, fluorescence microscopy and flow cytometry. The stability of this cell line for 20 passages following thawing has been verified.

What is the passage number of cells? ›

The degree of subculturing a cell line has undergone is often expressed as “passage number,” which can generally be thought of as the number of times cells have been transferred from vessel-to-vessel. A growing body of literature demonstrates passage number affects a cell line's characteristics over time.

Is it bad to passage cells two days in a row? ›

Yes it is harmful if you are trypsinizing your cells continuously after 24 hrs of splitting. It is advisable to do splitting after 48 hrs of splitting.

How many times can you passage primary cells? ›

Moreover, multiple passages can increase susceptibility of the primary culture to microbial contamination. Generally for primary cultures, passaging primary cells in between 2 to 5 are suitable for maintaining the genetic and phenotypic properties.

What counts as passaging cells? ›

What is subculture? Subculturing, also referred to as passaging cells, is the removal of the medium and transfer of cells from a previous culture into fresh growth medium, a procedure that enables the further propagation of the cell line or cell strain.

How often should cells be passaged? ›

Depending on the cell type, most adherent cells need to be passaged when they are 70-90% confluent, that is, when they cover 70-90% of the culture container surface.

How many passages after thawing cells? ›

Allow 3 or 4 passages after thawing a new vial of cells. Since contamination can drastically alter transfection results, cell cultures and media should be routinely tested for biological contamination (see Biological Contamination), and contaminated cultures and media should never be used for transfection.

How many cells should be counted? ›

At least 106 cells/ml are required for accurate counting. Mix the cell suspension sample thoroughly. Using a pipet, immediately transfer 20 µl to the edge of one side of the coverslip to fill one chamber of the hemocytometer.

What is the cell limit? ›

Total number of characters that a cell can contain. 32,767 characters.

What is the split ratio for cell passage? ›

As a general guide, from a confluent flask of cells: 1:2 split should be 70-80% confluent and ready for an experiment in 1 to 2 days. 1:5 split should be 70-80% confluent and ready for an experiment in 2 to 4 days. 1:10 split should be 70-80% confluent and ready for sub-culturing or plating in 4 to 6 days.

What is a limit cell size? ›

Cell size is limited by a cell's surface area to volume ratio. A smaller cell is more effective and transporting materials, including waste products, than a larger cell. As a cell grows, its volume increases more quickly than its surface area.

What is the maximum passage number for HEK293? ›

With proper handling and maintenance, HEK293 cells can typically be passaged between 20 and 50 times before experiencing significant changes in their growth characteristics or genetic stability.

What is the difference between passage number and generation number? ›

Passage number: Page 2 It is the number of times that the culture has been sub-cultured. Generation number: It refers to the number of doublings that a cell population has undergone. It must be noted that the passage number and generation number are not the same, and they are totally different.
